Details

ANX Sepharose 4 Fast Flow (high sub) is a weak anion exchanger, with a different selectivity compared with traditional ion exchangers. It is optimized for the separation of larger proteins Weak anion exchanger with different selectivity compared with traditional ion exchangers. Applicable for separation of high molecular weight proteins. Developed in co-operation with leading large-scale pharmaceutical manufacturers. High chemical stability allows for well proven CIP and sanitization protocols. The hydrophilic nature of the base matrix ensures low levels of non-specific binding leading to low levels of host cell-derived impurities in the elution pool. ANX Sepharose Fast Flow (high sub) is part of the Sepharose Fast Flow ion exchange platform, which has been the industrial standard for ion exchange chromatography during recent decades. ANX Sepharose 4 Fast Flow (high sub) is a weak anion exchanger resin with tertiary amine groups attached to the base matrix Sepharose 4 Fast Flow via ether linkage and a hydrophilic spacer arm. The diethylaminopropylgroup is coupled in a mode that precludes formation of quaternary groups, which are typically seen on traditional DEAE media, resulting in a truly weak anion exchanger. The base matrix, Sepharose 4 Fast Flow, is a highly cross-linked, 4% agarose derivative with high chemical and physical stability and broad separation range. ANX Sepharose 4 Fast Flow (high sub) has high chemical stability, allowing well proven cleaning-in-place (CIP) and sanitization protocols. As member of the BioProcess resin range, ANX Sepharose 4 Fast Flow (high sub) meets industrial demands with security of supply and comprehensive technical and regulatory support. BioProcess resin: Yes Ligand: Diethylaminopropyl Particle size, d50V: ~90 µm Ion Exchanger Type: Weak anion exchanger Matrix: cross-linked agarose, 4%, spherical Ligand Coupling Method: Ether linkages and hydrophilic spacer arm Ionic Capacity: 0.13-0.18mmol Cl-/ml medium pH stability, operational: 3–13 pH stability, CIP: 2–14 pH ligand fully charged: Below 9 Pressure/flow characteristics: ≥200 cm/h at <0.1 MPa in a XK 50/60 column with 5 cm diameter and 25 cm bed height (at 20°C using buffers with the same viscosity as water). Exclusion limit (Mr) (globular proteins): ~3 × 10⁷ Storage: 4 to 30°C, 20% Ethanol Chemical stability: Stable in commonly used aqueous buffers: 1.0 M NaOH, 20% and 70% ethanol, 8 M urea, 8 M guanidine HCl, 1M acetic acid, aqueous buffers -1.0 M NaOH, - 20 and 70% Ethanol Certificate of Analysis: Yes
